Laboratory reports contain a wealth of information about your peptide's quality, but interpreting these documents requires understanding the underlying metrics. This guide will help you extract meaningful insights from peptide verification reports.

HPLC Purity Data

Retention Time: The time it takes for the peptide to pass through the HPLC column. This value helps identify the peptide and should be consistent for a given peptide under standardized conditions.

Peak Area Percentage: The main peak area as a percentage of total integrated peak area. This is the primary measure of purity — a 98% peak area means 98% of the detected material is the desired peptide.

Impurity Peaks: Smaller peaks in the chromatogram represent impurities. These may include deletion sequences, truncated peptides, or degradation products.

Mass Spectrometry Data

Observed Mass: The molecular weight measured by the mass spectrometer. This should closely match the theoretical (calculated) molecular weight.

Mass Accuracy: The difference between observed and theoretical mass, typically expressed in Daltons or parts per million (ppm). High mass accuracy increases confidence in identity confirmation.

Charge States: In electrospray mass spectrometry, peptides appear at multiple charge states. The pattern of these peaks provides additional confirmation of molecular identity.

Additional Quality Metrics

Net Peptide Content: The actual peptide mass as a percentage of total vial contents. This accounts for counter-ions (typically acetate or TFA salts), moisture, and residual solvents.

Endotoxin Levels: Reported in endotoxin units per milliliter (EU/mL). Lower values indicate cleaner products suitable for sensitive biological assays.

Amino Acid Analysis: Confirms the correct ratio of amino acids in the peptide, providing another layer of identity verification.
